Coventry City line up against West Ham's U-21s team, with Wycombe Wanderers and Northampton Town completing the group stage line-up.
The 16 groups of four teams each comprise an academy side with the top two from the group qualifying for the first knockout round of 32 teams, and culminating in a showpiece final at Wembley Stadium on April 2.
The new look competition will also have an enhanced prize fund with an expected £1.95m up for grabs, including £10,000 per win and £5,000 per draw during the Group Stage and £100,000 for the eventual winner. Barnsley beat League Two side Oxford United 3-2 at Wembley in front of a crowd of more than 59,000 in last season's final.
